Identification of Novel Blood Biomarkers among Data-Driven Parkinson’s Disease Clusters in Asian Populations

Meeting: 2024 International Congress

Abstract Number: 107

Keywords: Parkinson’s

Category: Parkinson's Disease and Lewy Body Dementia

Objective: The aim of this study is to explore the correlation between emerging serum biomarkers and subtypes of Parkinson’s disease (PD).

Background: PD is a complex neurodegenerative disorder characterized by significant heterogeneity. Subtype identification and biomarkers exploration are the top priorities in the field of PD clinical research. However, the knowledge of biomarkers in PD subtypes remains markedly limited.

Method: The study enrolled subjects from the Early Parkinson’s disease Longitudinal Singapore (PALS) cohort. Six novel blood biomarkers including Neurofilament light chain protein (NFL), phosphorylated tau 181(p-tau181), a-synuclein, Glucose, Amylin and Ubiquitin carboxy-terminal hydrolase L1 (UCHL-1) were measured and analysed by the Kruskal–Wallis nonparametric test across three identified clusters.

Results: Two hundred and six early PD patients, consisting of 43 patients in cluster A, 98 patients in cluster B and 65 subjects in cluster C were recruited. The severe PD cluster has significantly higher levels of NFL, UCHL-1 and Glucose (cluster A B C:16.3, 14.3, 12.5, p=0.003; 2.5 1.8 0.0 p=0.007; 98.0, 94.0, 90.0, p=0.032 respectively) in the Kruskal–Wallis test. UCHL-1 remains significant in the Generalized linear mode after adjustment of age of diagnosis (p=0.02).

Conclusion: Our findings suggest that a blood-based panel including NFL, UCHL-1 and Glucose could be a useful prognostic tool for discerning the severity of PD patients. Further validation of these novel biomarkers is imperative through studies involving larger sample sizes. Additionally, future longitudinal investigations, which consistently monitor changes in these serum biomarkers over time, could offer valuable insights.
