Objective: To evaluate the association between MT-ATP6 gene polymorphisms, oxidative stress, and biomarker alterations in PD patients from Indian populaiton
Background: Mitochondrial dysfunction and oxidative stress are key contributors to Parkinson’s disease (PD) pathogenesis. Variations in the mitochondrial ATP synthase subunit 6 (MT-ATP6) gene may impair oxidative phosphorylation, resulting in excessive reactive oxygen species (ROS) generation and altered biochemical biomarkers.
Method: Clinically diagnosed PD patients and age- and sex-matched healthy controls were recruited. MT-ATP6 gene polymorphisms were identified using targeted sequencing. Oxidative stress was assessed by measuring intracellular ROS levels. Biomarker analysis, including uric acid (UA) and dopamine (DA) levels, was performed using enzyme-linked immunosorbent assay (ELISA). Statistical analyses were conducted to determine associations between genetic variants, oxidative stress parameters, and biomarker levels.
Results: PD patients demonstrated significantly elevated ROS levels compared to controls (p < 0.05). Specific MT-ATP6polymorphisms were more prevalent in PD patients and were significantly associated with increased ROS production. Furthermore, PD subjects exhibited reduced UA and DA levels, which correlated with higher oxidative stress and the presence of MT-ATP6 variants, indicating compromised mitochondrial function and dopaminergic neurodegeneration.
Conclusion: This study suggests that MT-ATP6 gene polymorphisms contribute to enhanced oxidative stress and altered biochemical biomarkers in PD. The combined assessment of MT-ATP6 variants, ROS levels, and UA and DA biomarkers may provide valuable insights into mitochondrial dysfunction and offer potential biomarkers for disease monitoring and therapeutic targeting.
