Background: Dayak Onion is a traditional plant used by the Dayak people (Kalimantan, Indonesia) to empirically treat various ailments. Various studies have shown this plant to be rich in compounds with potential as therapeutic candidates. This is relevant given that current therapy for Parkinson’s disease (PD) is limited to alleviating clinical symptoms. It is hoped that this research will reveal new hope for PD therapy
Method: In this work, we used a variety of bioinformatics approaches, including network pharmacology, molecular docking, and molecular dynamics, to investigate the possible therapeutic advantages and illustrate the fundamental mechanisms of dayak onion compounds against PD.
Results: The 10 target proteins can interact with all chemicals, according to molecular docking studies. Possible chemicals from dayak onion can be used to assess the ESR1, HSP90AA1, and AKT1 proteins in PD in silico. The chemicals methylanthraquinone, eutherine, and anthraquinone have the lowest docking scores against a number of target proteins when compared to levodopa, which is their control. Among the target proteins, Methylanthraquinone compound has the lowest docking scores (kcal/mol): HSP90AA1 (-11.18), AKT1 (-10.34), and ESR1 (-8.53). At -10.08 for HSP90AA1, -10.07 for AKT1, and -8.53 for ESR1, Eutherine has the lowest docking scores. Meanwhile, the Anthraquinone compound is known to have the lowest docking score against the HSP90AA1 protein (-10.58), AKT1 (-9.73) and ESR1 (-8.25).
Conclusion: Dayak onion contain methylanthraquinone, eutherine, and anthraquinone molecules that have a good pharmacokinetic profile and can bind to protein targets implicated in PD. As a result, they can be submitted for additional in vitro and in vivo studies.
