Effect of herbal medicine against Parkinson’s Diseases- An in silico studies

Objective: To evaluate the effect of the bioactive compounds of Gymnema sylvestre on treatment of Parkinson’s diseases.

Background: Neurodegenerative diseases, such as Parkinson’s and Alzheimer’s, are understood as occurring through multifactor pathophysiological mechanisms. More than 10 million people worldwide are living with PD. Herbal medicine (Gymnema sylvestre (GS)) have been used in the treatment of various problems including neurodegenerative diseases for a long time in folk system of medicine and the literature reported that some bioactive compounds are having the capacity to cross the blood-brain barrier and slow the progression of the diseases. The present study we to reported the bioactive compounds of GS on in silico enzymatic target studies and natural products as inhibitors for the treatment of Parkinson’s diseases.

Method: In this study we evaluated bioactive compounds using prediction of molecular properties and in silico docking studies, while comparing against 2 standard reference compounds.  Osiris analysis revealed that the compounds are non toxicity and good absorption parameters. silico docking studies by using PatchDock and FireDock, viewed by Acceryls discovery studio visualizer.  Finally the protein ligand complex was subjected to MD simulation stability assessment using Desmond software.

Results: The docking results as compared to the standards with four ( human adenosine receptor A2A, α-synuclein , Catechol-O-Methyltransferase, Monoamine Oxidase B ) proteins revealed similar binding energies, indicating that the bio active compounds are potential leads with the necessary pharmacological and structural properties to be drug candidates. The MD simulations up to 20ns exhibited a stable structure and ligand deviations (RMSD) less than 4.

Conclusion: results indicating that the GS bioactive compounds are potential drug candidates. However further studies need to confirm the same.
