Lack of association between LRRK2 G2385R and cognitive dysfunction in Korean patients with idiopathic Parkinson’s disease

Objective: We investigated whether the G2385R polymorphism in LRRK2, a risk allele for the development of idiopathic Parkinson’s disease (IPD) in East Asians, is associated with cognitive dysfunction in IPD.

Background: Aside from the glucocerebrosidase gene, the genetic risk factors for cognitive decline in IPD are controversial.

Methods: We recruited 299 IPD patients from 14 centers. Global cognitive function was assessed using the Mini-Mental State Examination (MMSE). IPD with cognitive impairment was defined as an MMSE Z score below -1.0 standard deviation. The MMSE score was adjusted by age at study entry and years of education.

Results: In univariate analysis, cognitive impairment in IPD was associated with older age at study entry, older age at IPD onset, longer IPD duration and depression, but not with the genotype of LRRK2 G2385R. In the multivariate analysis, IPD duration and depression were significant risk factors for cognitive impairment in IPD. In the linear regression analysis of the association between MMSE Z scores and IPD duration, there was no significant difference depending on the genotype of LRRK2 G2385R.

Conclusions: In conclusion, the genotype of LRRK2 G2385R may not be associated with cognitive dysfunction in IPD.
