Anti-parkinson effect of natural L-DOPA containing drug Mucuna pruriens against the rotenone induced rat model of Parkinson’s disease

Objective: The nanoherbaceutical formulation comprising natural L-DOPA from Mucuna pruriens against rotenone caused Parkinson’s disease in rats to be prepared and evaluated.

Background: Parkinson’s disease (PD) is an age-related neurological disorder and gradual degeneration of dopaminergic nigrostriatal neurons that results in cognitive impairment and activity. Rich source of natural 3-(3,4-dihydroxyphenyl)-l-alanine (L-DOPA), Mucuna pruriens has demonstrated anti-parkinson behavior without exhibiting any untoward effect.

Method: Rotenone was administered subcutaneously for PD induction. Two behavioral analyzes were conducted on rats, i.e. hang and catalepsy study evaluated. Brain homogenate has been used to respectively test the metabolic, antioxidant, pro-inflammatory, and inflammatory mediator.

Results: Nanoherbacterial therapy demonstrated increased levels of antioxidant levodopa (93 pmol / mg protein) like CAT (63.4%), SOD (69.5%), GPx (85.6%), and lower levels of MDA (79/4%) and nitrite in rat brain nigrostriatal tissue relative to PD. It also strengthened the neurobehavioral traits by growing the hanging period as contrasted with group rats caused by rotenone. It also significantly reduced pro-inflammatory cytokines like TNF-α (65.6%), IL-1β (57.6%), IL-6 (68.4%); inflammatory mediators like iNOS (64.3%), MPO (45.6%) and NF-kB (84.5%), respectively.

Conclusion: We may conclude that Mucuna pruriens alleviates oxidative stress and physiological defects in rats and has the potential of neuroprotecting Parkinson’s disease.
