Histone deacetylase 4 is regulated by microRNA 128 in SH-SY5Y cells of Parkinson’s disease

Objective: To explore the mechanisms and regulating effects of microRNA 128 in Parkinson’s disease (PD).

Background: PD is the second common progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons in the substantia nigra. It is currently understood that the pathophysiology of PD is not clear. Up to now, there is still no effective diagnosis and treatment. MicroRNAs are a class of small RNA molecules implicated in post-transcriptional regulation of gene expression during development. Recent studies show that microRNAs are playing an important role in the pathophysiology of PD. It has been reported that the overexpression of miR-128 could moderate neuronal responsiveness, suppresses motor activity, and alleviates motor dysfunction associated with Parkinson’s–like disease. Furthermore, Histone deacetylase 4 (HDAC 4) also play a principal role in neurodegenerative diseases. We hypothesized that microRNA 128 could regulate the expression of HDAC 4 in an in vitro cell model of PD.

Methods: HDAC 4 was selected as the target gene of microRNA 128 in SH-SY5Y cells treated with rotenone. To explore the changes of HDAC 4 expression levels in SH-SY5Y cells with over-expression and lower-expression of microRNA 128.

Results: In SH-SY5Y cells, enhanced expression of microRNA 128 could down-regulate HDAC 4 and reduce the expression of alpha-synuclein, increased neuroprotection to rotenone. Blocking of microRNA 128 function increased the expression of HDAC 4 and alpha-synuclein.

Conclusions: Our data imply that elevated levels of microRNA 128 may play a neuroprotective role in PD by downregulating HDAC 4.
