The pivotal difference in gut microbiota between multiple system atrophy and Parkinson’s disease

Objective: The knowledge gap between microbiome dysbiosis and pathogenesis of multiple system atrophy (MSA) remains.

Background: Mounting evidence has found significant alterations in the gut microbiome and aberrant metabolic pathways associated with various neurodegenerative diseases, such as Parkinson’s disease (PD). However, controversial results were noticed in MSA.

Method: Participants of MSA, PD and healthy controls (HCs) were enrolled at the Department of Medicine, National Taiwan University Cancer Center, Taipei, Taiwan. Participants went home with a fecal sample collection kit and were instructed to return a fecal sample at the next visit. Stool samples will be stored at -80℃ prior to further analyses. The 16sRNA sequencing using V3-V4 region was performed by Miseq. The sequencing results will be analyzed using the bacterial 16s rRNA database SILVA. After constructing the OUT table, a taxonomy table was made. Bacterial species difference analysis between the fecal samples were compared, including alpha diversity assay, beta diversity assay, linear discriminant analysis (LDA), and function prediction analysis by the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis.

Results: Overall, 26 MSA, 66 PD, and 26 HC were enrolled in 2020-2021. Eubacteriaceae is significantly associated with MSA. The LDA characterized four other microbiomes in MSA. The KEGG pathway enrichment analysis of the gut microbiota in three groups identified that pathways of fatty acid biosynthesis, DNA replication, replication_recombination and repair proteins were significantly associated with MSA.

Conclusion: We found key bacterial species associated with MSA and summarized our preliminary results regarding gut microbiota and their enriched pathways in MSA. However, mechanistic studies of microbiota-and-metabolites association in MSA are still lacking.
