Catecholamines interrelation in plasma of early diagnosed treatment-naive patients with Parkinson disease

Objective: To investigate the interrelation of catecholamines in the blood plasma of healthy people and PD patients.

Background: The study “START-1” was funded by Investment and Venture Fund of the Republic of Tatarstan and Fund for the Promotion of the Development of Small Forms of Enterprises in the Scientific and Technical Sphere.

Methods: The analysis of blood samples was performed on the base of the Institution of Russian academy of Science, Institute of Biology and Development by high pressure liquid chromatography with gas detection. Regression analysis was done in the program Statistica 12. Results of biochemical analysis were received from 45 patients with early stages of Parkinson disease and 25 healthy volunteers comparable by age and sex.

Results: Multiple regression analysis showed the differences in the correlations’ profile of healthy patients and PD ones. Healthy volunteers had marked correlation of DOPA concentration with the dopamine (DA) (p=0.013) and noradrenaline (NA) (p=0.014). Regression analysis showed that more than 84% of the concentration variability of NA and DA were explained by the DOPA level. At the same time DA and NA have had significant strong negative correlation (r=0.67, p=0.004), and DOPAC have no interrelations with other catecholamines. In all PD patients DOPAC level correlated with the DA concentration (p<0.04). Regression analysis showed that 62% of the concentration variability of DOPAC in patients with shaking form (SF) of PD and 78% of the concentration variability of DOPAC inpatients with rigid form (RF) of PD could be explained by the DA level. This may indicate the higher degradation level of DA in patients with PD. In RF of PD patients no other correlations were found. In SF of PD patients negative correlation between DA and DOPA was found (p=0.047). It may indicate the higher level of DA production in these patients like in healthy volunteers. At the same time SF of PD patients as healthy people had positive correlation of NA and DOPA levels (p=0.05)

Conclusions: There are differences in the interrelations of catecholamines in patients with different forms of PD. The cause of such changes is unclear. It is supposed that in SFof PD there is some compensatory mechanism intervened, while in ARF of PD this process is not activated, even suppressed.
