Effect and Mechanism of Shaoyao Gancao Decoction(SGD) in Treatment of Pain in Parkinson’s Disease Based on Network Pharmacology and Molecular Docking

Objective: Based on the method of network pharmacology, this paper discusses the potential effects and mechanisms of SGD in the treatment of pain in PD.

Background: Pain is one of the common non motor symptoms of PD. In China SGD is commonly used in the treatment of various pain, including neuralgia, joint pain, cancer pain and so on.

Method: The active components in White Paeony Root and liquorice root were selected as candidate compounds by the TCMSP. And the target information of White Paeony Root and liquorice root was obtained by PubChem and sea database. The target information of PD pain was obtained by GeneCard, OMIM and PharmGKB database. The common target gene information was obtained by comparing traditional Chinese medicine and disease targets. Using Cytoscape 3 8.0 to draw the drug active ingredient target network diagram with software. Using string to construct the PPI network, the core protein gene was obtained, and then the core protein was screened and sorted according to the degree value.

Results: The main active ingredients of SGD are quercetin, kaempferol, licochalcone a, beta-sitosterol and so on. AKT1, TP53, TNF and CASP3 are the main targets. Finally, Go enrichment shows that its biological process involves response to lipopolysaccharide, response to metal iron. The main results of molecular function enrichment are ubiquitin-like protein ligase binding. KEGG pathway enrichment mainly includes MAPK, AGE-RAGE, IL-17 and TNF signaling pathway. Similar to the previous main targets, the above pathways are involved in the regulation of cell survival mediated by inflammatory response, apoptosis, autoimmunity and growth factors. In addition, there is Toll like receptor signaling pathway, which is also the target of anti anxiety and depression drugs in the treatment of neuralgia.

Conclusion: SGD can protect dopaminergic neurons and improve skeletal muscle pain and central pain in PD by anti-inflammatory, anti apoptotic and regulating nutritional factors. The ubiquitin – like protein ligase binding can maintain immune homeostasis, inhibit apoptosis of spinal cord neurons, and improve neuropathic pain in PD.It can play the role of antioxidation, maintaining iron balance, and regulating Toll like receptor signal pathway to play the role of anti anxiety.
