Ameliorating effect of linalool on anxiety disorders in Parkinson’s mice and related mechanisms

Objective: The aim of this study was to verify the ameliorating effect of linalool(LIN) on anxiety-related affective disorders and the associated receptors in a MPTP Parkinson’s mouse model. It is hoped that new drugs can be explored to improve Parkinson’s disease anxiety disorder in the future.

Background: Above 85% in people with Parkinson’s disease will have anxiety-related psychiatric symptoms. At present, there is no specific treatment for non-motor symptoms, so it is of great significance to explore better drug treatment to improve the quality of life of PD patients.Non-motor symptoms are often ignored or misdiagnosed, although they seriously affect the quality of life and social function of PD patients, and clinically mainly rely on symptomatic treatment and comprehensive intervention, and there is no specific treatment. Therefore, it is particularly urgent to explore treatment strategies for PD mood disorders.

Method: We divided the PD mice into 4 groups(MPTP group;MPTP+LIN group;MPTP+LIN+PTZ group;WT group) of 10 mice after successful MPTP modeling (C57BL/6, 25 – 35 g, n = 30). An odor exposure chamber was used to inhale linalool for 30 minutes in the experimental group of mice for 7 days. Then, we performed behavioral experiments on all mice: pole climbing experiment, balance beam experiment, and EPM experiment to evaluate their exercise ability and anxiety disorder respectively.

Results: The average climbing time of the MPTP+LIN group is 6.69±0.99s, and that of the MPTP group is 7.09±1.41 sec.espectively. There was no significant difference in the average pole climbing time or number of slips between the MPTP+LIN group and the MPTP group(p>0.05). The results indicated that LIN had no significant improvement or inhibitory effect on the motor function of PD mice. However, the time in open arms in EPM was significantly increased in MPTP+LIN group (MPTP = 61.15 6.878 sec, MPTP+LIN = 93.44 8.672 sec, p = 0.0381) after linalool odor exposure. And this increasement can be reversed by antagonists GABA. Then, in the substantia nigra tissue of different groups of mice, it was found that dopaminergic neuronal damage was reduced in the LIN inhalation group.

Conclusion: Linalool inhalation can improve anxiety disorders in PD mice, but does not affect the motor function of mice. Linalool may act through the GABA receptor-associated olfactory pathway.
