Hesperidin ameliorates rotenone-induced Parkinson’s disease in mice via improving neuroinflammation and oxidative stress

Objective: The goal of the present study is to examine the neuroprotective property of hesperidin on rotenone-induced Parkinson’s disease in mice.

Background: The second most prevalent neurodegenerative ailment is PD (Parkinson’s disease) which affects both motor and non-motor capabilities. It is generally known that the neuropathological process that causes Parkinson’s disease (PD) begins in the gut before migrating to the Brain, supporting the significance of rotenone (environmental toxins). Hesperidin is a flavanone glycoside that has antioxidant and neuroprotective properties that may help people with Parkinson’s disease.

Method: All the mice were randomly divided into various groups and supplemented with hesperidin for 28-days. Weekly behavioral tasks were completed one hour following therapy. On day 28, all the mice were sacrificed and specific brain areas underwent immunohistochemistry examination and biochemical parameters.

Results: Rotenone-induced behavioural abnormalities such as working memory deficit, motor incoordination, and depression were reversed by hesperidin. Furthermore, the administration of hesperidin attenuates the oxidative stress/antioxidant parameters (lipid peroxidation, glutathione, superoxide dismutase) which were induced by rotenone and attenuated the acetylcholinesterase (AchE) activities in specific areas of the brain. Rotenone significantly increased the α-synuclein, TLR-4 (toll-like receptor 4,) iba-1 expression, and GFAP (glial fibrillary acidic protein), and while lowering the expression of tyrosine hydroxylase positive neurons (TH) in mice. This effect was alleviated by pretreating the mice with hesperidin.

Conclusion: This research supports hesperidin’s neuroprotective effects on rotenone-induced Parkinson’s disease (PD) via boosting anti-inflammatory pathways and antioxidant properties.
