5HTTLPR (44bp Ins/Del) polymorphism: Serotonergic subtype of Parkinson’s Disease

Objective: To find out the association of 5HTTLPR (44bp Ins/Del) polymorphism with PD neurotransmitter subtypes

Background: Parkinson’s Disease (PD) can be sub divided into serotonergic, non-adrenergic and cholinergic based on non-motor symptoms. Studies have shown the association of 5HTTLPR (44bp Ins/Del) polymorphism with PD risk. But factors determining the subtypes remain unknown. Therefore, current study aimed to find the association of 5HTTLPR (44bp Ins/Del) polymorphism with PD neurotransmitter subtypes

Method: PD patients (n=150) were recruited based on UK Parkinson’s Society Brain Bank Criteria. Polymorphism was assessed by PCR-RFLP method and its association with PD neurotransmitter subtype was analysed

Results: Among the PD patients serotonergic was most common subtype (66.44%), followed by cholinergic (16.78%) and nor-adrenergic (15.44%). Dominant (L/L vs L/S+S/S) (OR: 2.8, 95% CI: 1.3-5.9, p=0.009), recessive (L/L+L/S vs S/S) (OR: 5.1, 95% CI: 1.8-14.1, p=0.0007) and allelic models (L Vs S – OR: 2.6, 95% CI: 1.5-4.3, p=0.0001) of 5HTTLPR (44Ins/Del) were associated with risk for serotonergic symptoms

Conclusion: 5HTTLPR (44bp Ins/Del) gene polymorphism is associated with risk for PD as well as it may be contributing factor for serotonergic subtype of PD
