Exosomal α-synuclein in Multiple system atrophy and parkinson’s disease cause distinct synucleinopathies

Objective: In this study, we investigated the differences in exosomal α-syn between MSA and PD patients.

Background: Parkinson’s disease (PD) and multiple system atrophy (MSA) are both neurodegenerative diseases. The abnormal accumulation of α-synuclein (α-syn) in neurons and glial inclusions is the main pathophysiological feature of these diseases. Recently, the role of exosomal α-syn in the pathogenesis of MSA and PD had attracted considerable attention. However, the difference of exosomal α-syn from MSA and PD patients remains unclear.

Method: Plasma-derived exosomes were isolated from MSA and PD patients. Then, exosomes were injected into the unilateral striatum of mice using stereotaxic apparatus for 3 months. In this study, we tested the seeding activity of MSA and PD exosomes and their roles in the α-syn aggregation in the bladders of mice.

Results: The increased seeding activity (Figure 1) and α-syn aggregation in the bladders of mice (Figure 2) were found in mice injected with MSA exosomes compared to those injected with PD exosomes.

Conclusion: Collectively, our results suggest that exosomal α-syn from MSA and PD patients has a distinct effect on the pathology in vivo, and might provide a mechanistic explanation for the different clinical symptoms of MSA and PD patients.
