Derivation and characterization of Parkinson’s disease related pattern using functional magnetic resonance imaging

Objective: This study attempts to identify Parkinson’s disease-related pattern in Japan using functional magnetic resonance imaging (fMRI), and to characterize this pattern.

Background: Parkinson’s disease-related pattern has been well established in 18F-fluorodeoxyglucose positron emission tomography (FDG PET). In contrast, Parkinson’s disease-related pattern derived from fMRI was just analyzed for a diagnostic tool.

Methods: Fifteen controls and 15 patients with PD were enrolled in the present study. All subjects underwent structural and functional MRI. Amplitude of low frequency fluctuation (ALFF) was calculated for each voxel after several preprocessing steps, using fMRI. Parkinson’s disease-related pattern was derived from ALFF maps using scaled subprofile modelling/principal component analysis. We investigated association between expression level of Parkinson’s disease-related pattern and each clinical feature, such as age, disease duration and Hoehn-Yahr stage.

Results: Expression level of Parkinson’s disease-related pattern was significantly increased in PD group compared with control group (P = 0.01). There was negative correlation between Parkinson’s disease-related pattern expression and Hoehn-Yahr stage (P = 0.02, r = -0.59).

Conclusions: Parkinson’s disease-related pattern has been identified using fMRI, although this is not yet validated in different dataset. We plan to apply this pattern to the other cohort to test reliability.
