PP1α negatively regulates D2 receptor mediated cadmium induced motor dysfunctions: protective potential of Quercetin.

Objective: The present study has been focused to identify the molecular targets that involved in Cadmium-induced motor dysfunctions. At the same time, we also focused on the protective potential of quercetin in modulating such changes.

Background: Cadmium is a perpetual toxin with high occurrence in nature and is importantly considered as a potential risk factor in the development of neurological and neurodegenerative diseases. On the other hand, quercetin is considered as a potent antioxidant and have been reported as a strong neuroprotectant. The regulation of DARPP32 protein in striatum neurons strongly mediated by the protein phosphatase at its thr34 site which modulates the dopamine D2 receptor mediated motor functions. Therefore, we focused to find out the mechanism that involved in cadmium-induced motor dysfunction.

Method: Adult male rats of Wistar strain treated with cadmium chloride (5mg/kg, p.o) or quercetin (25mg/kg, p.o) or simultaneously with cadmium and quercetin under in-vivo experimentations. While differentiated pc12 cells were treated with the Cadmium and Quercetin individually and simultaneously in-vitro. The techniques used for assessing are western blotting, receptor binding, inhibitors study, and behaviour studies.

Results: The results of the present study exhibit that treatment with cadmium decreases the D2 receptors while quercetin exposure upregulated the receptors under western blotting and binding studies. Further using the inhibitors of DARPP32 levels decrease while PP1α levels increase in striatum area. The results of the behaviour studies shows the decrease in motor activity in cadmium treated group. While rats simultaneously treated with quercetin shows increased activity.

Conclusion: The results of the present study exhibit that the motor dysfunctions in Cadmium-induced group is mediated through PP1α regulator, which modulates the activity of DARPP32 through D2 receptors.
