Objective: To evaluate the neuroprotective effects of Globimetula cupulata in a rat model of Parkinson Disease (PD) induced by 6-hydroxydopamine (6-OHDA) injection in the striatum, and its potential to mitigate neuronal dysfunction and oxidative stress.
Background: PD is characterized by the gradual degeneration and impairment of neurons. Oxidative stress regulates neurodegenerative and neuronal dysfunction. Globimetula cupulata is recognized for its influence on motor dysfunction and its ability to alleviate oxidative stress in many neurological disorders, indicating a potential protective effect against neurological damage induced by 6-OHDA injection.
Method: Wistar rats were randomly assigned to sham, lesion (6-OHDA), and three lesion groups treated with extract of Globimetula cupulata at different doses for 3 weeks. Then, neurotoxin 6-OHDA was microinjected into the left striatum to induce PD, and motor behavior was assessed. Parameters including oxidative stress and inflammation were analyzed in striatal and midbrain homogenates.
Results: Pretreatment with Globimetula cupulata significantly enhanced motor function when compared with the 6-OHDA group. The oxidative stress was also improved following treatment with Globimetula cupulata, as evidenced by an increase in in levels of SOD, CAT, and GSH and a decrease in levels of MDA.
Moreover, Globimetula cupulata treatment reduced the expression of proinflammatory cytokines TNF-α and IL-6, indicating its anti-inflammatory effects.
Conclusion: Globimetula cupulata attenuates motor impairment generated by 6-OHDA, enhances oxidative stress, and diminishes inflammation. The data indicate that Globimetula cupulata may be a viable therapy option for Parkinson’s disease.
