miRNAs from blood serum as an early diagnostic biomarker for Parkinson’s disease: A study in an animal model

Objective: This work aims to elucidate miRNA-based biomarkers for the early stages of Parkinson`s Disease diagnosis in a chemically induced PD rat model.

Background: The burden of environmental factors and ageing brings various health issues, especially neurodegenerative diseases such as Parkinson`s disease. PD is a multifactorial disorder pathologically characterized by losing dopaminergic neurons in the mid-brain region, particularly in substantia nigra and pars compacta. Despite clinical examination, diagnosing the disease in its early stages is still challenging. Biochemical diagnosis is necessary to evaluate patients and their disease status. In recent times, cell-free miRNAs have become a significant domain in disease diagnosis. miRNAs are a short stretch of non-coding RNAs that regulate gene expression post-transcriptionally.

Method: Based on the Literature survey and existing large-scale transcriptome datasets of PD vs Healthy control, we identified specific numbers of miRNAs to be differentially expressed. The expression of each miRNA is validated in the early days of treatment. The PD-induced model in rats withdraws the blood and separates the serum by a standardized protocol. Total RNA was converted to cDNA and validated a set of miRNAs.

Results: Computationally we narrow down miRNAs through R packages from PPMI data. We found a set of miRNAs that only express in an early stage of the disease. Furthermore, we validate a set of miRNAs: miR-214, miR 124, miR-34b, miR-34c and let-7b-5p by qPCR in the early days of PD-induced rat serum.

Conclusion: This study explores and validates a set of miRNAs that work as a biomarker for early Parkinson’s disease diagnosis.
