Relationship between non-motor symptoms and dopamine transporter activity in Parkinson’ s disease

Objective:

We investigated whether dopamine transporter single-photon emission computed tomography (DAT-SPECT) binding correlates to non-motor symptoms in patients with Parkinson’s disease (PD).

Background:

DAT-SPECT binding at striatal presynaptic terminals is associated with the loss of functional neuronal activity, and correlates with duration and severity of motor features in PD. Regarding relationship between non-motor symptoms and striatal DAT availability, the association of DAT-SPECT binding with apathy in untreated and non-depressed de novo PD patients has also been reported. In addition, anosmic patients with PD showed the decreased striatal [123I]β-CIT binding.

Methods:

Forty patients with PD were evaluated to DAT-SPECT, motor and non-motor functions by MDS-UPDRS I, II, III, Odor Stick Identification Test for Japanese (OSIT-J), REM sleep behavior disorder (RBD) Screening Questionnaire of Japanese version (RBDSQ-J), and Single-Question Screen (RBD 1Q).

Results:

 DAT binding had negative correlations with age, scores of MDS-UPDRS I, II and III (p < 0.01, 0.001, 0.05). Subcategories on MDS-UPDRS I; sleep, constipation and pain were significantly correlated with DAT binding (p < 0.01). There was not any correlation between scores of OSIT-J, or RBDSQ-J and DAT binding.

Conclusions:

DAT binding correlated not only motor symptoms but also to some non-motor symptoms in PD.
