Objective: The current study exemplified the therapeutic potential of solid lipid nanoparticles (SLN) of berberine (BE-SLN) against oxidative stress in nigrostriatal tissue and neurobehavioral characteristics in rotenone induced rat model of PD.
Background: Parkinson’s disease (PD) is an age-related neurological disorder and progressive degeneration of nigrostriatal dopaminergic neurons which resulting into cognitive impairment and movement. Berberine is a natural alkaloid with a bright yellow color isolated from the plant Coptis chinensis. Berberine has been shown to regulate glucose and lipid metabolism in vitro and in vivo.
Method: Double emulsion solvent displacement method was used for the preparation of BE-SLN. Rotenone was subcutaneously injected at a dose level of 2 mg/kg body weight to Swiss albino rats. Two behavioural analysis on rats i.e. hang and catalepsy test were performed. Brain homogenate were used to assess the biochemical parameters such as catalase, glutathione peroxidase (GPx), superoxide dismutase (SOD), malondialdehyde (MDA), Nitrites. Pro-inflammatory cytokines, inflammatory mediator and histopathology were performed.
Results: Our results showed that chronic rotenone treatment causes motor deficits, decreased rearing behavior, mitochondrial dysfunction, and oxidative stress in rat model. Surface methodology suggests the 178.4 nm particle size and 0.310 polydispersity index for BE-SLN. Treatment with BE-SLN, significantly (P<0.05) upregulated the level of antioxidant parameters including catalase (52.5%), SOD (43.5%), GPx (57.6%) and reduced the level of MDA (60.3%) and nitrite of nigrostriatal tissue of rat brain. In addition, BE-SLN administration improved the neurobehavioral features by increasing hanging time as compared to rotenone induced group rats. It also significantly reduced the pro-inflammatory cytokines such as TNF-α (53.4%), IL-1β (43.4%), IL-6 (48.6%); inflammatory mediators like iNOS (53.5%), MPO (32.3%) and NF-kB (64.5%), respectively.
Conclusion: These findings revealed that BE-SLN administration alleviates oxidative stress and physiological abnormalities in rats and has a neuroprotective potential to treat PD.
