Electric stimulation of the nucleus subthalamicus is able to modulate prepulse inhibition. A pilot study in Parkinson´s disease

Objective: To evaluate if prepulse inhibition (PPI) is influenced by electrical stimulation in the Nucleus subthalamicus (STN) of patients with Parkinson´s disease (PD).

Background: PPI can be used as a measurement of sensorimotor gating important for filtering situation relevant information. PPI is similarly detected in all mammals making it possible to compare results of animal- and human studies. It is known that dopamine influences PPI. PPI deficits are found in neuropsychiatric disorders like schizophrenia or PD showing disturbances within the dopaminergic system. Deep brain stimulation (DBS) in the STN is able to improve motor symptoms in PD. Conceivably electric stimulation of the STN could influence PPI.

Methods: Patients with Parkinson´s disease underwent DBS of the STN. PPI was tested under two conditions: no stimulation and with best stimulation setting. Prepulse (85dB SPL) was given 30ms, 60ms and 100ms before presentation of the pulse (103db SPL).

Results: The PPI of the patients was altered by the best configuration setting of DBS.

Conclusions: PPI can be modulated by electrical stimulation of the STN, suggesting that sensorimotor gating can be influenced by deep brain stimulation in this area.
